Anti-inflammatory Effect of an Ecklonia cava Ethanol Extract in Macrophage via Inhibition of the NF-¥êB/MAPK Signal Pathways

Abstract
Ecklonia cava is a brown alga that evidences antioxidant, diarrhea and anticoagulant effect. However, the molecular mechanisms underlying its anti-inflammatory effects remain poorly understood. In the current study, the effect of Ecklonia cava ethanol extracts (ECEE) on lipopolysaccharide (LPS)-induced cytokine expression in RAW264.7 cells was investigated. Our results indicate that ECEE inhibits LPS-induced inducible nitric oxide (iNOS) and cyclooxygenase (COX)-2 in a concentration-dependent manner. Also, pre-treatment with ECEE significantly inhibited the expression of inflammatory cytokines, such as tumor necrosis factor-¥á (TNF-¥á) and interleukin-1¥â (IL-1¥â). Moreover, phosphorylation of mitogen-activated protein kinases (MAPKs) and nuclear translocation of nuclear factor-kappa B (NF-¥êB) were strongly suppressed by ECEE in LPS-stimulated RAW264.7 cells. These results indicate that ECEE has an anti-inflammatory effect in RAW264.7 macrophage through inhibition of the NF-¥êB/MAP kinase signal pathways.
